In the past, I have used the sticky side of a spacer to remove the unnoticed lint and dust specks from the mat of a partially sealed mat package. I would open a side and then spread the package enough to reach in and pluck the little boogers out with a thin spacer.

The other day I discovered a tool that works better then the spacer. A long thin painting knife. I had just used the painting knife to separate a double mat and it had a little bit of atg stuck to it. The atg had lost most of it's tack, but still picked up the speck with ease.

I was in and out of the mat in no time.
 
Jana uses 1/4" wide strips of matboard with a little sticky on them to get her specks. I am sure she will tell you better how she makes them.

I have this adversion to touching any of the artwork, even if it is the top of a mat with sticky. I fear residue. I also have very small, dry fingers and if it can be reached with one of these instruments, I can get my finger.

I usually unseal 3 sides and blow the speck out, then I gradually lay the glass back down while still blowing across the artwork, until the glass is back in place. That usually works for me . . . of course not on chalk art with no fixative. I would have to put all the pieces of chalk back together.
